As with any private company (or public for that matter) when making projections based on speculative or projected numbers, models can be highly sensitive to the inputs. Even using the latest venture rounds doesn't necessarily reflect the truest value of a company - yes, in the end a company is worth what someone is willing to pay for it, but the limited information available isn't enough for buyers of shares on a private exchange like SharesPost to make informed decisions, even if they are accredited investors. I think they under-estimate Twitter's longer term margins. Could be as high as 60%, in which case you could double the valuation.
